2009, NutritionCitation Excerpt :Caco-2 cells were cultured in 24-well plates at a density of 5 × 104 cells/mL. After an 18-h stabilization period in medium without fetal calf serum, the cells were treated with various concentrations of Na-butyrate (2.5–20 mM) alone or combined with carnitine (10 mM) for 48 h. For the final 2 h of culture, 3H-thymidine was added (2 μCi/ml) to monitor changes in DNA synthesis, as previously described [23,35]. Results are expressed as percentage of 3H-thymidine incorporation compared with control wells (medium alone).

Stress kinase pathways are essential in the regulation of cytokine-induced growth modulation of enterocytes [18–20]. Since the intracytoplasmatic part of the IL-17 receptor contains no death domain allowing to associate apoptosis inducing adapter molecules, contrary to various membrane receptors of the TNF-family, it is not so surprising that IL-17 is devoid of pro-apoptotic effects.

Similar to NF-κB, mitogen activated protein kinases (MAPKs) are also activated in response to multiple stimuli, including exposure to cytokines, growth factors, and bacteria [15–17]. The aim of this study was to examine the effect of adenosine on TNF-α-induced NF-κB signaling and IL-8 gene expression in IECs.
