In this regard, experimental studies that have evaluated colonic tumorigenesis in mouse models and human cancer cell lines have mainly shown a protective effect of estrogen, and both pro- and anti-proliferative activity of testosterone [11–13]. Observational studies of the association between use of hormone replacement therapy and colorectal cancer risk in postmenopausal women have shown inverse associations with ever use of combined estrogen-progesterone therapy and with current use of estrogen therapy alone [14].

However, there are conflicting reports in the literature on the chemopreventative potential of this donor, even when tested in the same model [51,52]. The observation that neither naproxen nor TBZ could produce the same reduction of aberrant crypt foci formation as seen with the hybrid molecule (ATB-346) is suggestive that both suppression of COX and delivery of H2S is required to achieve the observed effect.
