Isolation of an atl type-IV collagen cDNA clone using a synthetic oligodeoxynucleotide

Abstract

We have isolated a cDNA clone (pCIV-1-225) for the α1 subunit of basement membrane (type IV) collagen from a cDNA library made from Engelbreth-Holm-Swarm mouse tumor RNA. The cDNA library was screened with synthetic oligodeoxynucleotides derived from published amino acid (aa) sequences (Schuppan et al., 1982). Nucleotide sequence data established the identity of our cDNA clone to encode an α1 type IV collagen. This clone contains 270 aa of the helical region and has three interruptions in the Gly-X-Y repeat unit.

    Although the significance of the triple‐helix interruptions is not absolutely clear, it can be assumed that they confer flexibility to the molecule and facilitate interaction with similar molecules or other macromolecular components of basement membranes. The complete amino acid sequence of the human α1(IV) chain was reported by Brazel et al. (1987) and Soininen et al. (1987), and for the mouse α1(IV) chain, it was reported by Nath et al. (1986), Wood et al. (1988), and Muthukumaran et al. (1989). The human α1(IV) chain is 1669 residues in length, with a putative 27‐residue signal peptide, a 145‐residue 7S domain, a 1268‐residue triple‐helical region, and a 229‐residue NC1 domain.
