Abstract
Background Alistipes is a class of gram-negative bacteria, which belongs to Bacteroidetes. Studies have indicated its protective effects against liver fibrosis, hypertension, and atrial fibrillation, while it’s implicated in the pathogenesis of colorectal cancer and psychological symptoms of depression. However, there is limited research examining its role in ulcerative colitis (UC).
Methods Comparing fecal samples from 30 UC patients with 6 healthy donors and 15 UC patients in active with remission to investigate the clinical impact of Alistipes on colitis by using 16S-rDNA high-throughput sequencing. 30 male BALB/c mice were randomly divided into three groups: healthy control (Control), DSS-induced colitis (DSS) and Alistipes_finegoldii intervention (DSS+AF). Colitis was induced by administering 2.5% DSS in drinking water. Control and DSS groups received PBS, while the DSS+AF group received Alistipes_finegoldii (108-9 CFU/mL) by gavage for seven consecutive days. The anti-inflammatory effect was evaluated by using disease activity index (DAI), colon shortening, histological damage, and expression of pNF-κB/NF-κB.
Results In the UC and HD groups, the Firmicutes/Bacteroidetes ratio, a marker of gut dysbiosis, was 2.67 and 3.14. LEfSe analysis revealed a significant enrichment of Alistipes in the HD group, with an LDA Score of 4.05 (P<0.0001). Compared to UC patients, Alistipes was enriched in the gut microbiota of healthy individuals (IDDF2024-ABS-0335 Figure 1 (A). LEfSe analysis between UC and HD). Conclusions Alistipes may be a potential indicator of clinical remission in UC. Alistipes_finegoldii has shown effectiveness in improving experimental colitis.
