A new enzymatic method for determination of serum choline-containing phospholipids
Abstract
A new enzymatic method is presented for the determination of serum choline-containing phospholipids with a combined enzymatic method using phospholipase D (from Streptomyces species), choline oxidase (from Arthrobacter species) and peroxidase. The method is reproducible, and the results correlate well with those obtained by the conventional digestion method (Hoeflmayr, J. and Fried, R. (1966) Med. Ernaehr. 7, 9–10). The method affords better specificity, requires a smaller quantity of the sample and shorter time than those previously reported, and has excellent precision.

Electrochemical analysis of total phospholipids in human serum for severe sepsis diagnosis
2024, TalantaElectrochemical analysis of total phospholipids was performed for the diagnosis of sepsis. The influence of electrode materials on the analysis of the chromogenic substrate was analyzed using Au, graphite, and pyrolyzed carbon electrodes. The total phospholipid analysis based on electrochemical analysis with pyrolyzed carbon was used for diagnosis of sepsis using sera from healthy volunteers, systemic inflammatory response syndrome (SIRS), and severe sepsis patients. The analysis results using the optical measurement and the electrochemical analysis were compared for the serum samples from sepsis patients and healthy controls. Additionally, the interference of human serum on the optical measurement and electrochemical analysis was estimated by signal-to-noise (S/N) calculation. The assay results of the levels of other biomarkers for sepsis (C-reactive protein and procalcitonin) and the total phospholipid levels obtained using the optical measurement and electrochemical analysis methods were statistically similar. Finally, the mortality of patients, indicated by the results of the total phospholipid assay performed using the electrochemical analysis of the patient samples collected daily (1, 3, and 7 day(s) after admission to hospital), was compared with the patient mortality assessed via conventional severity indexes, such as the SOFA and APACHE Ⅱ scores. The 28-day survival rate was estimated by Kaplan–Meier survival analysis based on the total phospholipid level of patient samples that were obtained after 1, 3, and 7 day(s) from hospital admission.
Absolute quantification of bovine lactadherin to screen the anti-rotavirus activity of dairy ingredients
2023, Journal of Dairy ScienceThe anti-rotavirus components in breast milk and infant formulas play an important role in the prevention of rotavirus infection. The present study examined whether the levels of phospholipids and bovine lactadherin, which are the major components and proteins of the milk fat globule membrane complex, are useful indices of the anti-rotavirus activity of dairy ingredients used in infant formulas. We compared the anti-rotavirus activity of 2 types of dairy ingredients enriched in the milk fat globule membrane complex: high-fat whey protein concentrate (high-fat WPC) and butter milk powder (BMP), using 50% inhibition concentration (IC50) and linear inhibition activity to determine levels of solid contents, total proteins, phospholipids, and bovine lactadherin. Here, we developed a quantification method using full-length isotope-labeled proteins to measure bovine lactadherin levels in these dairy ingredients. The evaluation of anti-rotavirus activity showed that the difference in IC50 was the smallest when the 2 dairy ingredients were compared at the bovine lactadherin level, among other indices in this study. Additionally, no significant difference was observed between the inhibition linearity of 2 dairy ingredients when evaluating only bovine lactadherin levels. These results indicated that the level of bovine lactadherin was more strongly associated with anti-rotavirus activity than the level of phospholipids. Our results suggest that bovine lactadherin levels can be used to estimate the anti-rotavirus activity of dairy ingredients and can be a criterion used in selecting ingredients for infant formulas.
Formulation and characterization of liposomes

They are gaining significant consideration in pharmaceutical, cosmeceutical, as well as food applications due to their unique properties, such as biocompatibility, biodegradability, flexibility for surface modification, versatility in encapsulating a wide range of bioactive molecules and releasing them in a particular fashion as desired and, moreover, ease of preparation and scale up. This chapter begins with a discussion of the mechanism of liposome formation followed by liposomal formulation by conventional methods with their features and complications, novel formulations along with their pros and cons, utility, and possibility to scale up. The chapter also discusses various physicochemical characterizations to assess the quality of liposomes. In addition, the spotlight is also put on the in vivo fate of liposomes after oral administration.
Pre- and postnatal polychlorinated biphenyl exposure and cognitive and behavioral development at age 45 Months in a cohort of Slovak children
2022, ChemosphereEvidence of associations of pre- and postnatal exposure to polychlorinated biphenyls (PCBs) with cognitive development beyond early childhood is inconsistent. A previous report from this cohort observed adverse associations between early life PCB exposures and infant Bayley scores at age 16 months. The present study examines pre- and postnatal PCB exposures in relation to both behavior and cognitive development at age 45 months. Participants were 472 mother-child pairs residing in an area of eastern Slovakia characterized by environmental contamination with PCBs, which resulted in elevated blood serum concentrations. PCB-153 and PCB-118 concentrations were measured in maternal and in infant 6-, 16-, and 45-month serum samples. At age 45 months, children were administered five subtests of the Wechsler Preschool and Primary Scale of Intelligence (WPPSI-III), and mothers completed the Child Behavior Checklist (CBCL). Negative binomial and multiple linear regressions were used to estimate PCB—CBCL and PCB—WPPSI-III subtest score associations, respectively. Pre- and postnatal levels of PCB-153 and PCB-118 were not associated with cognitive performance on the WPPSI-III in this cohort. There was some suggestion that higher postnatal PCB concentrations were associated with more sleep problems and feelings of depression and anxiousness.
Wet-chemically synthesis of SnO<inf>2</inf>-doped Ag<inf>2</inf>O nanostructured materials for sensitive detection of choline by an alternative electrochemical approach
2021, Microchemical JournalHere, a reliable electrochemical sensor prove was fabricated by SnO2-doped Ag2O nanorods (NRs) decorated glassy carbon electrode (GCE) to detect choline (CA) in the buffer phase. The wet-chemically synthesized SnO2-doped Ag2O NRs were characterized by field emission scanning electron microscopy (FESEM), X-ray diffraction (XRD), energy-dispersive X-ray spectroscopy (EDS) and X-ray photoelectron spectroscopy (XPS) techniques respectively. SnO2-doped Ag2O NRs was layered as a film of NRs onto a GCE using Nafion adhesive to obtain the working electrode of the choline (CA) sensor. The proposed sensor is calibrated by using the concentration of CA versus measured current, which is found linear over a concentration range of CA (0.1 nM ~ 0.01 mM) defined as a dynamic range (LDR) for choline detection. Using LDR slope and the active surface of GCE (0.0316 cm2), the choline sensor sensitivity (95.1 µAµM-1cm−2) is calculated. The limit of detection (LOD) and limit of quantification (LOQ) of the proposed CA sensor are calculated using the signal-to-noise ratio at 3, and the obtained values are 95.9 ± 4.8 pM and 319.67 ± 5 pM respectively. The sensor reliability measuring parameters (response time, stability and reproducibility) are found satisfactory. In the end, the choline sensor with SnO2-doped Ag2O NRs/GCE is performed preciously in the electrochemical analysis of various real bio-samples to validate its real-time applicability.
Diagnosis of severe sepsis using phospholipids enzymatic assay based on cyclic voltammetry
2021, Enzyme and Microbial TechnologyIn this work phospholipid quantification was carried out using an enzymatic assay based on cyclic voltammetry of the condensation product of N-ethyl-N-(2-hydroxy-3-sulfopropyl)-3,5-dimethoxyaniline sodium salt (DAOS) and 4-aminoantipyrine (4-AP) with a graphite electrode. For the optimization of electrochemical measurement for the product, electrochemical properties such as the electrochemical window, double layer capacitance (Cdl) and electron transfer rate (kapp) were analyzed for a graphite-electrode and Au-electrode. The phospholipid enzymatic assay based the on electrochemical measurement using the graphite electrode was applied to the diagnosis of sepsis for sera from healthy volunteers (n = 16), patients with systemic inflammatory response syndrome (SIRS, n = 16) and severe sepsis patients (n = 24). Finally, the phospholipid quantification results from the electrochemical measurement were statistically compared with the conventional method based on optical density measurement.
