The IBS-D model was established in mice as previously reported, with certain adjustments [20, 29]. Briefly, mice were anesthetized with ether, their colons were instilled with 0.1 mL TNBS dissolved in 50% ethanol (50 mg/mL) 2 cm from the anus, and mice were held upside down for 30 s to prevent TNBS from leaking out.

Animal studies reveal that stress can induce dysfunction in the epithelial barrier, (Lennon et al., 2013; Øines et al., 2012; Cameron and Perdue, 2005; Chen et al., 2017), alter gastrointestinal motility (Williams et al., 1988; Larauche et al., 2009), influence visceral pain (Greenwood-Van Meerveld and Johnson, 2017), and aggravate colitic disease (Lennon et al., 2013; Reber et al., 2006; Deng et al., 2016; Çetinel et al., 2010; Gue et al., 1997; Koh et al., 2015a; Mackos et al., 2016; Matsunaga et al., 2011; Reber et al., 2008; Schultz et al., 2015; Veenema et al., 2008; Xu et al., 2008). For instance, colitis alters stress responses (Reichmann et al., 2015, 2013; Kresse et al., 2001), and both stress and depression can reactivate colitis (Ghia et al., 2009; Qiu et al., 1999).
