2022, International Journal of PharmaceuticsCitation Excerpt :In clinical trials of various malignancies, celecoxib was effective in reducing morbidity (Arber et al., 2006; Bertagnolli et al., 2006; Steinbach et al., 2000), and celecoxib has fewer gastrointestinal, renal and platelet side effects than other non-steroidal anti-inflammatory drugs (NSAIDs) (Goldstein et al., 2000; Kim et al., 2011; Lemmens et al., 2020). However, the higher doses required for chemoprevention may limit the use of celecoxib, as celecoxib doses have been associated with cardiovascular disease, myocardial infarction, stroke, or heart failure (Kerr et al., 2007; Psaty and Potter, 2006; Solomon et al., 2005; Wallace, 1997). Studies have shown that combining NSAIDs with other chemopreventive drugs can enhance efficacy and reduce the doses required for each drug, thereby reducing side effects caused by high-dose single-agent administration (Agarwal et al., 1999; Jalving et al., 2005; Reddy et al., 2006; Xiao and Yang, 2008; Zheng et al., 2007).
